General description
ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Each ZooMAb® antibody is manufactured using our proprietary recombinant expression system, purified to homogeneity, and precisely dispensed to produce robust and highly reproducible lot-to-lot consistency. Only top-performing clones are released for use by researchers. Each antibody is validated for high specificity and affinity across multiple applications, including its most commonly used application. ZooMAb® antibodies are reliably available and ready to ship when you need them.
Photosystem II protein D1 (UniProt: P83755; also known as EC:1.10.3.9, PSII D1 protein, Photosystem II Q(B) protein) is encoded by the psbA gene (Gene ID: 844802) in Arabidopsis thaliana (Mouse-ear cress). PSBA is a plastoquinone oxidoreductase that uses light energy to abstract electrons from H2O, generating O2 and a proton gradient subsequently used for ATP formation. It is a vital component of the PSII complex, facilitating the photochemical conversion of light energy into chemical energy during photosynthesis. Structurally, D1 forms a heterodimer with the D2 protein and is integral to the reaction center, where it plays a critical role in the binding of chlorophyll and the transfer of electrons. PSII D1 protein is a multi-pass membrane protein and is expressed in chloroplasts, primarily in the thylakoid membranes, where it undergoes rapid turnover in response to environmental stressors, such as high light intensity, which can induce photoinhibition. Dysregulation of D1 protein expression or mutations in the psbA gene can lead to impaired photosynthetic efficiency and has been implicated in various plant stress responses and adaptations. In higher plants, alterations in D1 expression have been associated with reduced growth and yield. In cyanobacteria, defects in D1 can lead to severe consequences for photosynthesis, including a loss of water-splitting ability and increased susceptibility to photodamage and, cell death under stress conditions. Recent studies highlight the significance of D1 in the maintenance of chloroplast integrity and its potential role in plant resilience to climate change. Research study suggested that expression of psbA is dynamically regulated at the translational level in response to light and PSII damage and, changes in mRNA secondary structure might represent a general mechanism for translational regulation of psbA and other plastid genes. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Immunogen
KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal half of Arabidopsis thaliana (Mouse-ear cress) PSBA.
Application
Quality Control Testing
Evaluated by Western Blotting with recombinant PSBA protein.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected recombinant Arabidopsis thaliana (Mouse-ear cress) PSBA.
Tested Applications
Enzyme Immunoassay: A representative lot of this antibody detected recombinant Arabidopsis thaliana PSBA in ELISA application.
Affinity Binding Assay: A representative lot of this antibody bound PSBA peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions
Clone 1J22 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects PSBA. It targets an epitope within 25 amino acids from the C-terminal half.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
