Take an appropriate amount of Ganmao Qingre granules, grind into powder, take about 0.8g, weigh accurately, transfer into a conical flask with a stopper, add 50 mL of a 30% ethanol solution, cover the stopper, and weigh them. Conduct ultrasonic treatment (250 W, 33 kHz) for 20 minutes, then cool down to room temperature. Weigh again, make up the lost weight with 30% ethanol. Shake well, filter, and stand for HPLC analysis.

Ganmao Qingre granule is a famous Chinese patent medicine. It is an external antidote, which has the effect of dispersing wind and cold, relieving external heat and clearing away heat. Puerarin is considered to be the main active ingredient of Ganmao Qingre granules.

The theoretical plate count for the described method is 8636, thus meets the requirement of the Chinese pharmacopeia 2020 stating NLT 4500. The repeatability for a 10 µg/mL puerarin in 30% ethanol standard solution was determined with an % RSD of 0.08.
The Limit of Detection (LOD) for puerarin is 0.05 µg/mL and the Limit of Quantification (LOQ) is 0.16 µg/mL. The range from 0.1 to 50 µg/mL is linear, with an R2 of 0.9998.
